Many tire manufacturers require scheduled rotations to keep the mileage warranty of the tire valid. Keeping your tires rotated will help all four tires wear at the same time, thus maintaining proper handling.
If you have a full size spare and wish to have that added to the rotation, simply install the spare to the right rear, and the tire that would have gone to the right rear should replace the spare with each rotation.
Note: Please note that some vehicles have staggered or directional tire size fitments and cannot be rotated in a traditional manner.
